A team of scientists from Japan’s Shibaura Institute of Technology has developed supercharged vitamin K analogues that could one day help the brain heal itself. These engineered compounds, which combine vitamin K with retinoic acid (a form of vitamin A), have shown a remarkable ability to regenerate neurons, offering fresh hope for treating diseases like Alzheimer’s and Parkinson’s.

What does “Supercharged” Vitamin K mean?

The term “supercharged vitamin K” refers to a scientifically enhanced form of natural vitamin K that has been modified to work faster, more efficiently, and more powerfully in the body, especially in the brain.
While regular vitamin K (like menaquinone-4, or MK-4) supports normal functions such as blood clotting and bone health, these newly engineered analogues have been chemically upgraded to improve their ability to:

  • Penetrate the blood-brain barrier (a natural protective shield that blocks many substances from entering the brain).
  • Stimulate neuron formation by activating specific brain receptors.
  • Remain stable and active for longer inside the body.

In short, supercharged vitamin K is not a supplement found on store shelves; it’s a lab-designed version created by scientists to unlock vitamin K’s full potential for brain regeneration and repair.

What’s the breakthrough?

Neurodegenerative diseases like Alzheimer’s, Parkinson’s, and Huntington’s cause neurons to die, leading to memory and movement issues. Current treatments manage symptoms but can’t reverse the damage. Researchers led by Professors Hirota and Suhara have developed supercharged vitamin K compounds three times more potent than natural vitamin K at stimulating new neuron growth, offering a potential breakthrough in brain regeneration.

How does vitamin K affect the brain?

Vitamin K is traditionally known for its role in blood clotting and bone health, but growing research points to its importance in brain protection. The Japanese team focused on MK-4, a form of vitamin K that influences neuron development. However, MK-4 on its own lacks the strength needed for use in regenerative therapies.
To address this, the researchers synthesized 12 hybrid vitamin K compounds by linking vitamin K molecules with retinoic acid or other chemical groups. Retinoic acid, derived from vitamin A, is known to encourage neuron formation. By combining the two, the team created supercharged vitamin K that merges the best features of both molecules.

The star compound: Novel VK

Among the 12 hybrids, one stood out. This particular compound, combining retinoic acid with a methyl ester side chain, triggered a threefold increase in neuron formation compared with natural vitamin K. It was named Novel VK.
In lab studies, Novel VK not only promoted neuron growth but also successfully crossed the blood-brain barrier, a critical advantage for any potential brain treatment. Once inside, it efficiently converted into MK-4 and maintained stability, meaning it stayed active longer in the body.
Dr. Hirota explained, “The newly synthesised supercharged vitamin K analogues demonstrated approximately threefold greater potency in inducing the differentiation of neural progenitor cells into neurons compared to natural vitamin K. Since neuronal loss is a hallmark of neurodegenerative diseases, these analogues may serve as regenerative agents that help replenish lost neurons and restore brain function.”

What’s the mechanism behind it?

Researchers found that supercharged vitamin K activates the brain receptor mGluR1, boosting stem cells to form new neurons. Novel VK binds more strongly than natural vitamin K and also triggers epigenetic changes that promote neuron growth and protection.

What could this mean for neurodegenerative diseases?

If these findings hold up in future studies, supercharged vitamin K could open a new era in treating neurodegenerative disorders. Instead of just slowing disease progression, these compounds might one day reverse damage by regenerating neurons and restoring lost brain functions.
Dr. Hirota shared his optimism: “Our research offers a potentially groundbreaking approach to treating neurodegenerative diseases. A supercharged vitamin K-derived drug that slows the progression of Alzheimer’s disease or improves its symptoms could not only enhance the quality of life for patients and their families but also reduce the societal burden of long-term caregiving.”
